Viruses
In comparison with caesium chloride and sucrose, OptiPrep™ provides major improvements in the purification of viruses. Recovery of infectivity is higher and particle:infectivity ratios are lower, sometimes by as much as 10-100x.
The loss of surface glycoproteins that often occurs, with retroviruses in particular, in sucrose gradients, does not occur with OptiPrep™ gradients.

Subcellular Membranes
Iso-osmotic gradients prepared from OptiPrep™ provide improved resolution of many subcellular membranes, including nuclei, mitochondria, ER, Golgi, TGN, plasma membrane domains, endosomes, peroxisomes and transport vesicles from both mammalian cells and from yeast.
Many applications employ the unique advantage of the ability of OptiPrep™ to form self-generated gradients; these reduce preparation time and improve reproducibility and resolution.

Lipoproteins and Proteins
Simple technology allows the purification of lipoproteins in 3 hours, rather than the 3 days required by routine sequential flotation; most add-on techniques can be performed without the need for dialysis. The high water activity of OptiPrep™ gradients is ideal for analysis of protein complexes that are unstable in other gradient media such as glycerol.
